Searches for NMN supplements in Malaysia have grown fast over the past year. They are sold for energy and healthy ageing, and the claims made for them range from careful to far-fetched. This guide explains what an NMN supplement is, how NMN becomes NAD+ in the body, and what human clinical trials and recent meta-analyses actually show, including where the evidence is still missing. It is written for formulators and brand owners, but the science applies to anyone weighing NMN.

What is an NMN supplement (nicotinamide mononucleotide)?

NMN stands for nicotinamide mononucleotide. It is a nucleotide: a molecule built from nicotinamide (a form of vitamin B3), a ribose sugar and a phosphate group. The body uses NMN as a precursor, a building block, to make NAD+ (nicotinamide adenine dinucleotide). [1]

An NMN supplement is a dietary supplement that supplies NMN as a powder, in capsules or tablets, or in sachets. Human trials have used β-nicotinamide mononucleotide (β-NMN). [2][3] NMN also occurs naturally in foods such as vegetables and fruit, but the trials tested supplement doses.

Is NMN just vitamin B?

No. NMN is made from nicotinamide, which is a form of vitamin B3, but NMN itself is not a vitamin. It is one step closer to NAD+ than nicotinamide is. Nicotinamide riboside (NR) is another NAD+ precursor sold as a supplement. Human trials have tested both NR and NMN. [1]

Why NAD+ matters

NAD+ is a coenzyme found in every living cell. It plays a central role in cellular energy metabolism, in the cell’s redox balance, and in signalling pathways linked to ageing. [4] Enzymes involved in DNA repair also use NAD+.

NAD+ levels in the body decline with age. This age-related decline is the reason for “NAD+-boosting” strategies, from lifestyle changes to supplements of NAD+ precursors such as NMN and NR. [1] In animal studies, raising NAD+ was often linked to better metabolic, mitochondrial and functional outcomes. [1] The key question for an NMN supplement is whether those effects carry over to people.

How NMN converts to NAD+: the pathway and how NMN supplementation can boost NAD+

Cells make NAD+ through several routes. In the salvage pathway, nicotinamide is turned into NMN, and NMN is then converted into NAD+ in a single enzyme step. This is why NMN is described as a direct precursor to NAD+. Once NMN is absorbed, the same final step applies. Human clinical trials confirm that NMN supplementation by mouth raises levels of NAD+ in the blood:

  • In a 60-day double-blind clinical trial of the efficacy and safety of NMN in 80 healthy middle-aged adults, 300, 600 or 900 mg of NMN a day raised blood NAD+ in every NMN group compared with placebo. Levels were highest at 600 and 900 mg. [2]
  • In a 14-day trial in 32 overweight or obese adults aged 55–80, 1,000 mg of NMN once or twice a day produced dose-related increases in blood NAD+. Very little unchanged NMN was lost in the urine. [5]
  • A 2026 systematic review of 33 human studies found that oral NMN and NR consistently raised NAD-related metabolites. [1]

So the first step works: NMN in humans raises NAD+, and higher doses of NMN gave larger increases. Whether increasing NAD+ leads to health benefits is a separate question, covered next.

NMN or NR?

Nicotinamide riboside (NR) is the other NAD+ precursor widely sold as a supplement. The 2026 systematic review found that both NMN and NR, taken by mouth, consistently raised NAD-related metabolites and were generally well tolerated, and that their effects on function and health outcomes were similarly mixed. [1] The trials cited here do not show that one is better than the other.

Potential benefits of NMN: effects of NMN in human trials

Many of the NMN supplement benefits you see online come from animal studies, where NMN has shown promising effects. Here is what controlled clinical trials and meta-analyses in people show, area by area.

Healthy ageing and longevity

A 2026 systematic review of 113 studies (33 in humans) concluded that NAD+ boosting has clear biological activity, but that its clinical effectiveness for anti-ageing effects or wellness outcomes is still inconclusive. In human studies, effects on function, metabolism and blood vessels were mixed and often null. [1] No trial has shown that NMN extends lifespan in people.

Blood sugar and metabolic health

Two meta-analyses found no clear metabolic benefit. A 2024 meta-analysis of 8 randomised trials in 342 adults found that 250–2,000 mg a day for up to 12 weeks did not improve fasting glucose, fasting insulin, HbA1c, insulin resistance (HOMA-IR, a measure of insulin sensitivity) or blood lipids. [6] A 2026 meta-analysis of 15 trials also found no significant effect on body weight, BMI, fasting glucose, HbA1c or lipids. [7]

Muscle and physical performance

A 2025 meta-analysis of randomised trials in adults over 60 found that NMN did not improve muscle mass, grip strength, gait speed or the chair-stand test. [8] A 24-week trial in 14 older men with diabetes and reduced physical performance found that 250 mg of NMN a day was safe but did not improve grip strength or walking speed. [9] One 60-day trial in healthy middle-aged adults did report longer six-minute walking distance at 300–900 mg a day. Several of its authors are from industry. [2]

Blood pressure

This is the clearest signal so far, and it is small. A 2026 meta-analysis of 10 trials (349 participants) found NMN lowered resting diastolic blood pressure by about 2 mmHg. Systolic pressure did not change overall but fell by about 4 mmHg in people aged 60 and over. The authors call the evidence preliminary, so NMN may lower blood pressure slightly, but this needs confirming. [10] A second 2026 meta-analysis also found a slight fall in diastolic pressure. [7]

Energy, skin and the brain

NAD+ is central to cellular energy production, so NMN products are often marketed for energy, skin and brain health. Claims that NMN improves energy, skin or memory within weeks are not backed by the meta-analyses above. Personal before-and-after stories cannot separate the effect of NMN from placebo. If an NMN product makes these claims, ask for the clinical trial behind them.

Potential risks and side effects: is NMN safe to take daily?

In trials lasting up to 24 weeks, taking NMN daily has been safe and well tolerated:

  • A 2026 meta-analysis of 15 trials (250–2,000 mg a day, 14 days to 24 weeks) found that NMN did not increase overall, serious or withdrawal-related adverse events, and did not raise the liver enzymes ALT or AST. [7]
  • In a 4-week double-blind trial of supplementation with NMN in 31 healthy adults aged 20–65, 1,250 mg a day caused no changes beyond normal physiological variation and no severe adverse events. Most of its authors are from industry. [3]

NMN dosage: how much NMN do people take?

Human trials have used 250 to 2,000 mg a day, for 14 days to 24 weeks. [7] In the dose-comparison trial, blood NAD+ rose most at 600 and 900 mg a day. [2] These are doses of NMN used in research, not a recommended dosage. There is no evidence for different doses by age group. The dose on a finished product must follow the rules of the market where it is sold.

NMN and NAD+: which should you take?

NAD+ itself is about twice the size of NMN. A 2026 systematic review reported that oral NMN and NR consistently raise NAD-related metabolites in people, and found no outcome trials of NAD+ drips or injections for anti-ageing or wellness. [1] We compare the options in NMN vs NAD+.
